G9a, in complex with the highly homologous GLP, is the major histone H3 lysine-9 (H3K9) methyltransferase for euchromatin and can also dimethylate p53K373, a modification correlating with levels of inactive p53. This latter result, along with the fact that G9a and GLP are overexpressed in a variety of human cancers suggests G9a/GLP inhibition as a potential anti-cancer therapy, especially for tumors expressing wild-type p53.
