SMYD2 has been described as a histone H3 (H3K4, H3K36), p53 (p53K370) and Rb (retinoblastoma; RbK860) lysine methyltransferase, although RBC finds this recombinant SMYD2 preparation to be far more active in vitro with recombinant histone H4 than with histone H3, an H3(1-21) peptide, an H4(1-21) peptide, core histones or recombinant p53. SMYD2 is frequently overexpressed in esophageal squamous cell carcinoma (ESCC) and SMYD2 knockdown inhibits the proliferation of SMYD2-overexpressing ESCC cell lines.
