Human DOT1L, and its homologs in other species, are unique among the histone lysine methyltransferases in not containing a SET domain and in targeting a specific residue in the histone H3 globular domain (lysine-79; H3K79).
Inappropriate recruitment of DOT1L by MLL-fusion proteins is implicated in the development of leukemias.
